Components(final concentrations)

2M Tris base

1 M acetate

100mM EDTA

H2O


To make 1 liter(usually kept as stock and diluted when used)

242g Tris base

57.1 mL glacial acetic acid (17.4M) N.B: GLACIAL ACETIC ACID WILL BURN YOU! USE IN HOOD AND AVOID ALL CONTACT!  If in doubt about how to handle glacial acetic acid speak to your mentor first!

100 mL of .5M EDTA(pH 8.0)

H2O up to a liter


(1X TAE pH 8.1 is 40mM Tris, 20mM acetate and 2mM EDTA)

use at 1X

50x TAE

EDTA: P/N: OMNI-PUR 4050

TRIS BASE: P/N: SIGMA T1503-500G

GLACIAL ACETIC ACID: P/N: SIGMA-ALDRICH 695084-2.5L
